چکیده

Human tumors and tumor cell lines were analyzed for the presence of mRNA coding for transforming growth factors a and ß(TGF-a and -ft) and the epidermal growth factor receptor. TGF-a mRNA was not detect able in hematopoietic tumor cell lines but was found in a variety of solid tumor cells, particularly carcinomas. Many of the tumors that contained TGF-a mRNA also expressed high levels of epidermal growth factor receptor mRNA. The concentration of TGF-a in the media of several tumor cell lines did not necessarily correlate with TGF-a mRNA levels, as a substantial fraction of TGF-a can remain cell associated. The levels of TGF-/3 mRNA in tumor cell lines and tumor specimens were variable, but higher in tumors than in the adjacent normal tissues.

Transforming growth factor-beta increases steady state levels of type I procollagen and fibronectin messenger RNAs posttranscriptionally in cultured human dermal fibroblasts.

Transforming growth factor-beta (TGF beta), when injected subcutaneously into newborn mice, induces a rapid fibrotic response, stimulates chemotaxis, and elevates the rates of biosynthesis of collagen and fibronectin by fibroblasts in vitro. We explored the molecular mechanisms of TGF beta-mediated stimulation of collagen and fibronectin synthesis in cultured human foreskin fibroblasts. TGF bet...

Proteomics and microRNA profiling in cardiac fibrosis

Cardiac fibrosis is characterized by the deposition of extracellular matrix (ECM) and mediated by cardiac fibroblasts via the renin-angiotensin system (RAS) and transforming growth factor beta (TGFβ) signaling pathways. Recognition of TGFβ by fibroblasts leads to myofibroblast differentiation as well as induction of ECM protein expression. An imbalance between proand anti-fibrotic signals resul...

EGF, basic FGF, and TGF beta-1 messenger RNA production in rabbit corneal epithelial cells.

The polymerase chain reaction (PCR) was used to demonstrate that rabbit corneal epithelial cells produce messenger RNAs coding for epidermal growth factor (EGF), basic fibroblast growth factor (FGFb), and transforming growth factor beta-1 (TGF beta 1) ex vivo and in primary culture. EGF, FGFb, and TGF beta 1 mRNAs were detected in central and peripheral ex vivo epithelial tissue in wounded and ...

Transforming growth factor-beta stimulates inorganic phosphate transport and expression of the type III phosphate transporter Glvr-1 in chondrogenic ATDC5 cells.

Members of the transforming growth factor (TGF)-beta family are important regulators of skeletal development. In this study, we investigated the effect of TGF-beta1 on inorganic phosphate (Pi) transport and on expression of the type III Pi carriers Glvr-1 and Ram-1 in murine ATDC5 chondrocytes. TGF-beta1 induced a selective, dose- and time-dependent increase in sodium-dependent Pi transport in ...

Effect of high glucose on peritoneal mesothelial cell biology.

OBJECTIVE This study reviews evidence that implicates high glucose (HG) in the pathogenesis of peritoneal fibrosis and proposes mechanisms potentially involved in the HG-induced peritoneal fibrosis that is observed in long-term peritoneal dialysis (PD) patients. DESIGN Selected Western literature is reviewed, examining the effect of HG on rat or human peritoneal mesothelial cell (HPMC) biolog...
